In-vitro and bioinformatic studies of bioactive compounds from Oceanimonas sp. JM-AZM31 and Lysinibacillus fusiformis JM-AZM37 of sponge-associated marine bacteria from a mangrove habitat in Southeast Sulawesi
The ongoing quest for novel therapeutic agents has directed attention toward bioactive compounds derived from sponge-associated bacteria. This study focuses on sponge symbiont bacteria from the mangrove ecosystems in Tanjung Tiram, Southeast Sulawesi, which have not yet been reported for their potential antibacteria, anti-inflammatory, antioxidant, and anti-diabetic activities. The screening of marine bacterial isolates was performed using a series of assays: disc diffusion method to assess antibacterial activity, protein denaturation to assess anti-inflammatory properties, DPPH free radical scavenging to evaluate antioxidant capacity, and α-Glucosidase inhibition for anti-diabetic activity, followed by in silico validation. Two promising strains, identified through molecular techniques were designated as Oceanimonas sp. JM-AZM31 and Lysinibacillus fusiformis JM-AZM37. Initial bioactivity screening revealed significant potential: The bacterial isolates JM-AZM31 and JM-AZM37 demonstrated broad-spectrum antibacterial activity against both Gram-positive pathogens (Bacillus cereus, Staphylococcus aureus, Staphylococcus epidermidis) and Gram-negative pathogens (Escherichia coli, Salmonella typhimurium). JM-AZM31 exhibited an anti-inflammatory inhibition rate of 76.9 ± 2.90 %, antioxidant activity of 80.3 ± 1.02 %, and anti-diabetic activity of 84.9 ± 0.49 %. Similarly, JM-AZM37 showed anti-inflammatory activity of 71.6 ± 1.85 %, antioxidant activity of 76.9 ± 0.03 %, and anti-diabetic activity of 83.2 ± 1.27 %. Further analysis using GC-MS identified five significant compounds, which were examined through in silico molecular docking. Results indicated that n-hexadecanoic acid, DL-proline, 5-oxo-, and cis-vaccenic acid showed high binding affinities to specific therapeutic targets, suggesting strong potential as biotherapeutic agents. This current inquiry concentrates on the therapeutic potential of marine bacteria from mangrove ecosystems as a source of bioactive compounds, positioning Oceanimonas sp. JM-AZM31 and L. fusiformis JM-AZM37 as promising candidates for developing new biotherapeutic treatments.
Detection of Resistance Gene from Aeromonas hydrophila Isolated from Catfish Farming in Jambi
Aeromonas hydrophila is one of the causative agents for Motile Aeromonas Septicemia (MAS) in catfish. MAS treatment is generally carried out by administering antibiotics. This study aimed to determine the antibiotic resistance profile of A. hydrophila and detect the presence of resistance genes. The samples are archived A. hydrophila isolates isolated from catfish ponds in the district of Muara Jambi, Bungo, Jambi Province, and Merangin total of 18 isolates. Testing the antibiotic resistance profile was carried out using the disk diffusion method against tetracycline, oxytetracycline, ampicillin, amoxicillin, ciprofloxacin, enrofloxacin, and streptomycin. The detected resistance genes were ampC, qnrA, qnrB, and qnrS. The results showed A. hydrophila isolates were resistant to tetracycline (100%), oxytetracycline (94.42%), ampicillin (77.70%), and amoxicillin (77.72%) and still sensitive to ciprofloxacin (83.32%), enrofloxacin (100 %), and streptomycin (100%). The ampC gene was found in 8 out of 14 isolates resistant to beta-lactam antibiotics (57.14%). The qnrA and qnrS genes were found in 3 out of 4 isolates resistant to quinolone antibiotics (75%), while qnrB was found in 1 isolate (25%). A. hydrophila isolated from catfish ponds in Jambi develops resistance to antibiotics and resistance genes found in isolates resistant to antibiotics.
Chemicals Analysis of Drilled Ground Waters in Moa Island, Maluku, Indonesia
Moa Island is one of the remote Indonesian Islands that face the difficulty in accessing clean water, especially during the dry season. One way to get clean water is by drilling for groundwater. The drilled groundwater in Kaiwatu Village on Moa Island, Southwest Maluku Regency, Maluku Province, was analyzed for its content to obtain primary data. Primary data related to the quality of drilled groundwater sources includes chemical, physical, and microbiological parameters. The results of chemical parameters obtained from the test results were <0.01 mg/L fluoride, total chromium, nitrite, nitrate, iron, manganese, zinc, cyanide, lead, <0,001 mg/L cadmium, 17.9 mg/L hardness, sulfate 147.55 mg/L, pH 7.50, and organic substances 2.8 mg/L. The physical parameter analysis showed a turbidity level of 3.71 NTU, a temperature of 26.8 °C, and a color of 5 TCU. The microbiological parameters obtained by the analysis were 0/100 mL. Primary data was then compared with secondary data, namely environmental health quality standards and air health requirements from Minister of Health Regulation No. 32 of 2017 for chemical and physical parameters, as well as water quality requirements and monitoring from Minister of Health Regulation 416/MENKES/IX/1990 for microbiological parameters. The analysis results using a comparison of primary and secondary data showed that all parameters contained in the primary data had values below the safe limits set by the requirements in the secondary data.
Antibiotic Resistance Patterns of Staphylococcus aureus in Chicken Farms: Implications for Health Resilience in Indonesia
Health resilience refers to an individual's or a community's ability to withstand and recover from various health challenges and crises. Antibiotic resistance poses a significant threat to health resilience due to its impact on the effectiveness of antibiotics. The urgency of the issue of multidrug bacterial resistance in public health is a concern. The phenomenon discussed in this paper is related to the environment that becomes resistant to the antibiotic Staphylococcus aureus which causes high morbidity and mortality in animals such as chickens. In this study, there were three levels of antibiotic resistance based on clear zone media culture, namely resistance, intermediates, and sensitivity with each antibiotic totaling nine chicken cloacal swab samples. The purpose of this study was to determine the pattern of the significance of antibiotic resistance in S. aureus in three chicken farms in West Java Province (Bogor, Parung, and Sukabumi). This study used secondary data from previous studies and analyzed with nonparametric statistical methods using the Kruskal Wallis Test (One Way ANOVA). The results of the analysis showedalmost all gave a resistant response characterized by the absence of a clear zone in bacterial culture media treated with the same antibiotic in all chicken cloaca swab samples taken from farms in Bogor, Parung, and Sukabumi. Thus, resulting in the same resistance pattern. This indicates the pattern of the resilience of resistance in West Java is the same because the ecological conditions and living standards of the community are almost the same. These results may illustrate important concerns for the possibility of other regions in Indonesia having similar patterns of antibiotic resistance, so antibiotic use must adhere to appropriate guidelines to combat further antibiotic-resistant strains.
Identification of Antibiotic Resistance Pattern and Antibiotic Resistance Gene of Staphylococcus aureus from Broiler Chicken Farm in Sukabumi and Cianjur Regency, West Java Province, Indonesia
Abstract Broiler chicken is one of the most common forms of protein in Indonesia. Sukabumi and Cianjur are two of the province of West Java’s top producers of broiler chickens. Farmers nonetheless use antibiotics for prevention, treatment, and growth enhancement. The objective of this investigation is to detect antibiotic resistance and the related resistance gene. The cloacal swab samples were determined to be Staphylococcus aureus by biochemical and molecular assays. Against the positive S. aureus strains, the antibiotics tetracycline, ampicillin, gentamicin, nalidixic acid, ciprofloxacin, erythromycin, and chloramphenicol were evaluated. The colony’s obvious inhibitory zone was evaluated in accordance with the 2018 Clinical and Laboratory Standard Institute recommendations (CLSI 2018). After extracting the DNA from resistant strains, the polymerase chain reaction (PCR) method was employed to identify the resistance gene. The total number of S. aureus isolates from both regions was 26, with 14 coming from Sukabumi and 12 from Cianjur. The vast majority were resistant to the vast majority of antibiotics used in broiler farms including several medicines. These samples carried the genes blaTEM, gyrA, tetA , and ermB for antibiotic resistance.
Antibiotic Resistant Pattern and Resistant Gene Identification of Staphylococcus aureus from Chicken Farm in Bogor
Chicken is one of the important protein source in Indonesia. Moreover, the largest population of chicken layer and poultry in Indonesia is known situated at West Java province with Bogor manicipality as the main producer. The aims of this study were to determine the antibiotic resistance pattern of Staphylococcus aureus isolated from poultry and layer farm in Bogor. The study also identified gene encoded the resistance. Cloacal swab samples were collected from chicken broiler and layer farm in Bogor manicipality. The samples were then cultured in Mannitol Salt Agar (MSA) medium to obtain S. aureus. Suspected colony was then confirmed by biochemical test. Positive strains were tested against several antibiotics and the diameter of clear zone arround of colony was interpreted based on Clinical and Laboratory Standard Institute. Furthermore, the DNA from resistant strains were then extracted, followed by detection of the resistance gene by using polymerase chain reaction (PCR) method. A total of 14 isolates of S. aureus were positive from poultry farm, and 15 isolates from layer farm. Most of all were resistant to tetracycline, ampicillin, oxytetracycline, erythromycin and nalidixic acid. On the other hands, several strains were sensitive to gentamycin and chloramphenicol. The study showed 28 isolates out of them were multi-drug resistant. Resistant gene such as blaTEM, gyrA and tetA were also identified in some isolates except for ErmB gene which was found in isolates originated from poultry farm. In conclussion, S. aureus in both farm showed mostly multi-drug resistant to several antibiotics which were supported by identification of resistant gene among isolates.
Antibiotic resistance profile of Staphylococcus aureus, Streptococcus spp. and Klebsiella spp. isolated form chicken farm in Bogor, Sukabumi, and Cianjur, West Java
Abstract West Java province has largest population of chicken poultry, with Bogor, Sukabumi, and Cianjur has highest chicken population. Farmers used antibiotics for prophylaxis and therapy to maintain the production. However, extensive use of antibiotic increased the number of antibiotic resistant bacteria. Staphylococcus aureus, Streptococcus spp, and Klebsiella spp are the example of flora normal in chicken that affected with abusive use of antibiotic. The aim of this study was to determine the antibiotic resistance profile of S. aureus, Streptococcus spp., and Klebsiella spp. isolated from cloacal swab of chicken poultry in 3 regions. Total of 320 samples were collected and the positive number of S. aureus , Streptococcus spp., and Klebsiella spp were 61, 8, and 58 isolates respectively. The result of antibiotics susceptibility test showed that S. aureus was resistant to ampicillin (98%), erythromycin (95%), nalidixic acid (93%), tetracycline (92%), oxytetracycline (90%), enrofloxacin (69%), and ciprofloxacin (56%). Streptococcus spp. was resistant to tetracycline (100%) and doxycycline (87.5%). Klebsiella spp. was resistant to erythromycin (100%), ampicillin (94.83%) oxytetracycline (93.10%), tetracycline and nalidixic acid (89.66%), enrofloxacin (86.21%), and ciprofloxacin (81.03%). S. aureus, Streptococcus spp., and Klebsiella spp. has a high level of resistance to antibiotics and most of the isolates were multi-resistant.
MP-7 Isolation of Antibiotic Resistance Gene from Staphylococcus aureus from Chicken Poultry Farm in Bogor West Java Indonesia
Staphylococcus aureus is an opportunistic pathogen in human and another animal species. It also associated with staphylococcosis, osteomyelitis, arthritis, and bumble foot disease in poultry. Antibiotics including penicillin, erythromycin and tetracycline are widely used for treating infection caused by S. aureus . Chicken poultry play important role in food supply as one of protein source in Indonesia. West Java province has largest population of chicken poultry, where Bogor has the largest population of chicken poultry with 19.75.850 chicken in 2018. In order to maintain the production, farmers used antibiotics for prophylaxis, growth promotor, and therapy. However, extensive use of antibiotic can be found in low income and developed country, caused the increasing number of antibiotic resistant bacteria. The aim of this study is to determine the antibiotic resistance pattern of S. aureus isolated from cloacal swab of chicken poultry in Bogor West Java Indonesia.
Ekstraksi Protein dari Escherichia coli BL21 Rekombinan Gen Mycobacterium tuberculosis dengan Variasi Waktu Inkubasi Induksi Isoprophyl-β-D-Thiogalactosidase (IPTG) dan Metode Lisis Sel
Bacillus Calmete-Guerin vaccine is no more effective in preventing tuberculosis. The vaccine developed now is vaccine recombinant using Mycobacterium tuberculosis gen cloned in Escherchia coli to produce protein which is used as antigen compound in vaccine. The protein produced an intercellular protein which is needed cell lysis process so that the protein could be extracted. The aim of this research was to investigate the incubation time, and the optimum cell lysis method in protein recombinant extraction. Recombinant E. coli BL21 was incubated for 4, 8, 12, and 16 hours, and followed by, freeze thaw, sonication, and beads vortex lysis method. SDS PAGE quantification showed that the sample with 12 hour incubation with freeze thawing lysis method had the highest protein recombinant concentration by 4.327.870,4 pixel.
